Cimicifuga rhizome extract induces apoptosis through caspase-3 activation in SCC 25 cells

Abstract
This study was undertaken to verify the induction and pathway of apoptosis in oral squamous carcinoma cells (SCC 25) using cimicifuga rhizome extract. In the present study, the WST-1 assay was used to detect the cell viability of cimicifuga rhizome extract on SCC 25 cells. Immuno-fluorescence and flow cytometry assays were used to detect cell apoptosis. The effect of cimicifuga rhizome extract on apoptosis-associated proteins (PARP-1, caspase-3 and caspase-9) were determined by western blot. We found that cimicifuga rhizome extract significantly inhibited cell viability in a dose-dependent manner in SCC 25 cells. Also, cimicifuga rhizome extract increased the sub G1 and changed the cytochrome c and AIF location. In addition, we observed that the caspase-3 signaling pathway played a key role in cimicifuga rhizome extract induced apoptosis. The results of the present study showed that cimicifuga rhizome extract promotes apoptosis via regulating the caspase-3 signaling pathway in SCC 25 cells.
